Background of Cell
The DF-1 cell line is spontaneously immortalized and can be propagated indefinitely. It has the typical cell morphology of fibroblasts. The DF-1 cells are reverse transcriptase negative, devoid of the endogenous gene sequences of derosarcomas and leukemia viruses, and non-tumorigenic. The DF-1 cell line is a globally recognized commercial cell line authorized by the United States Food and Drug Administration (FDA). It has been widely used in the propagation of avian viruses, the expression of recombinant proteins, the development of brain tumor viruses, and the production of animal and human vaccines. DF-1 cells are susceptible to and more apparent with chicken infectious bursal virus, reovirus, avian leukosis virus, roussarcoma virus, and Turkey herpesviruses. Viruses can replicate in DF-1 cells and cause observable cell lesions.
These cells are adherent-dependent cells and are typically cultured in a basic medium supplemented with fetal bovine serum. A 1:1 ratio of D/F12 medium to 10% fetal serum has been found to be optimal. After optimization and screening, an AEM medium suitable for DF-1 cell culture was determined. This medium is chemically defined and only requires 3% newborn bovine serum to support DF-1 cell growth. It can be used for serum-free virus inoculation and as a serum-free virus maintenance medium to support the proliferation of some viruses.
