CHO-WH1 Medium CHO-WM1 Medium is a culture medium specially designed for Chinese hamster ovary (CHO) cells. This medium supports the suspension culture and expression of recombinant proteins in CHO cells. CHO-WM1 Medium does not contain animal-derived components. It can support the growth of CHO cells in both the dihydrofolate reductase (DHFR) and glutamine synthetase (GS) systems (with additional supplementation of hypoxanthine and thymidine), and it does not contain phenol red to minimize the estrogenic effects of phenol red-like compounds.
1. Add 95% of the final volume of ultrapure water to the suitable container, add an appropriate amount of mediumand stir to dissolve, add ultrapure water to the final volume, and stir for 30 minutes to fully dissolve.
2. Adjust the pH to 6.85-7.15 with 1N sodium hydroxide or concentrated hydrochloric acid solution as needed.
3. Remove bacteria by filtration with a 0.2ym micron filter membrane, and the solution should be stored in thedark at 2-8°C. CHO-WM1 medium needs to be supplemented with L-glutamine before use.
4 Add L-alutamine 6mM final concentration (30m/ L) aseptically to the medium before use.
5. CHO-WM1 medium is free of hypoxanthine and thymine for use with the dihydrofolate reductase (DHFR)amplification system. For other use, add 10 ml/ L HT before use.
6. lf cell clumping occurs, add 1m/L anti-agglomeration agent. After this, the number of agglomerated cells canbe reduced by more than three times compared with that before use.
Note: lf you are using a batch feed. perfusion protocol. or if the fine material used is ammonia sensitive. considerusing a minimum level of L-glutamine.
Note: No surfactant (such as F-68)is required.
